Bioavailability Engineering for Detox & Cleanse Formulations

FRL applies evidence-based bioavailability strategies to optimize the exposure, stability, solubility, permeability, and delivery of selected active ingredients while considering formulation compatibility and regulatory requirements.

Bioavailability Engineering

Effect is dictated by exposure, not dose. FRL identifies the enhancement strategy based on the physicochemical properties of the active and the regulatory requirements of the target market.

  • Phospholipid complexation (phytosome-like technology) — increases the permeability of poorly soluble flavonolignans and polyphenols without interfering with any pharmacological enzyme inhibitors
  • Self-nano-emulsification systems (SNEDDS/SEDDS) and nanoemulsions — average droplet size D50 below 200 nm, determined by dynamic light scattering, prepared using high-pressure homogenization or microfluidization
  • Cyclodextrin inclusion complexation — solubility and taste-masking gains in a single step
  • Amorphous solid dispersions by hot-melt extrusion or spray drying, with physical stability confirmed by DSC and XRPD
  • Microencapsulation and enteric coating of probiotics and heat-sensitive actives, water activity maintained below the viability threshold for the specific strain
  • Screened out where mechanistically inconsistent: absorption enhancers working through Phase II enzyme inhibition and efflux transporters; these increase the drug level in blood by inhibiting the very pathways the product claims to enhance, expanding its potential herb-drug interaction profile.

Enhancement is proven, not presumed: INFOGEST 2.0 static digestion to establish bioavailability, Caco-2 monolayer permeability with TEER assessment, and when the commercial rationale warrants, a randomized crossover pharmacokinetic study reporting Cmax, Tmax and AUC relative to the unformulated reference substance.
